目的观察中药复方健脾解毒方对裸鼠皮下移植瘤Bax及Bcl-2 mRNA表达的影响。方法采用HCT8/V细胞制作裸鼠皮下移植瘤,将32只造模成功的裸鼠随机分为4组,每组8只。空白组予0.9%Na Cl溶液腹腔注射,化疗组予长春新碱腹腔注射,中药组予健脾解毒方灌胃,联合组予长春新碱腹腔注射和健脾解毒方灌胃。测量各组皮下移植瘤的瘤重及抑瘤率,光镜观察肿瘤组织病理情况,并采用Real-time PCR法测定Bax、Bcl-2 mRNA的表达。结果与空白组比较,化疗组、中药组、联合组瘤重明显降低,差异有统计学意义(P<0.01);与化疗组比较,中药组、联合组瘤重均降低,差异有统计学意义(P<0.01);相对于空白组,化疗组、中药组、联合组瘤体抑制率分别为22.84%、38.44%、51.53%,以联合组抑瘤效果最佳。与空白组比较,化疗组、中药组、联合组Bax mRNA表达水平显著升高(P<0.05,P<0.01),Bcl-2 mRNA表达水平均显著降低(P<0.01);与化疗组比较,中药组、联合组Bax mRNA相对表达量高于化疗组(P<0.05,P<0.01),中药组、联合组Bcl-2 mRNA相对表达量均低于化疗组(P<0.01)。结论健脾解毒方通过上调Bax mRNA表达,下调Bcl-2 mRNA表达,促进肠癌细胞的凋亡,且与化疗药具有协同作用。
Objective To observe the effect of Jianpi Jiedu decoction on the expression of Bax and Bcl-2 mRNA in nude mice xenografts. Methods HCT8 / V cells were used to make subcutaneous xenografts in nude mice. Twenty-two nude mice were randomly divided into 4 groups with 8 mice in each group. The blank group was intraperitoneally injected with 0.9% Na Cl solution. The chemotherapy group was injected with vincristine intraperitoneally. The Chinese medicine group was given Jianpi Jiedu Decoction intragastrically and the combination group was given vinbacracillin intraperitoneal injection and Jianpi Jiedu Decoction intragastrically. The tumor weight and tumor inhibition rate of subcutaneous tumor in each group were measured. The histopathology of tumor was observed by light microscope. The expression of Bax and Bcl-2 mRNA was detected by Real-time PCR. Results Compared with the blank group, the tumor weight of the chemotherapy group, the traditional Chinese medicine group and the combined group was significantly lower (P <0.01), and the tumor weight of the traditional Chinese medicine group and the combined group was significantly lower than that of the chemotherapy group (P <0.01). Compared with the blank group, the inhibition rate of the chemotherapy group, the traditional Chinese medicine group and the combined group tumor was 22.84%, 38.44% and 51.53% respectively, and the combination group had the best tumor inhibition effect. Compared with the blank group, the expression of Bax mRNA was significantly increased (P <0.05, P <0.01) and the expression of Bcl-2 mRNA was significantly decreased in the chemotherapy group, the traditional Chinese medicine group and the combination group (P <0.01) The relative expression of Bax mRNA in Chinese medicine group and combined group was higher than that in chemotherapy group (P <0.05, P <0.01). The relative expression of Bcl-2 mRNA in Chinese medicine group and combined group was lower than that in chemotherapy group (P <0.01). Conclusion Jianpi Jiedu decoction can promote the apoptosis of colorectal cancer cells by up-regulating the expression of Bax mRNA, down-regulating the expression of Bcl-2 mRNA and cooperating with chemotherapy drugs.
