目的观察mTOR及其信号通路分子在食管鳞状细胞癌和外周T淋巴细胞的表达。方法收集40例食管鳞状细胞癌术后癌组织、癌旁组织和外周T淋巴细胞,运用免疫组化、实时定量聚合酶链反应及流式细胞术检测mTOR、raptor及p-TSC2的表达量。结果癌组织mTOR和raptor表达量明显高于癌旁组织。临床Ⅲ~Ⅳ期mTOR蛋白的阳性表达率明显高于临床Ⅰ~Ⅱ期。患者外周血T淋巴细胞p-mTOR、p-TSC2的表达量高于正常人。结论食管鳞状细胞癌及外周T淋巴细胞中mTOR信号通路活化程度强于正常人。
Objective To observe the expression of mTOR and its signal transduction molecules in esophageal squamous cell carcinoma and peripheral T lymphocytes. Methods The expressions of mTOR, raptor and p-TSC2 in 40 cases of esophageal squamous cell carcinoma were detected by immunohistochemistry, real-time quantitative polymerase chain reaction and flow cytometry after cancer tissues, pericarcinomatous tissues and peripheral T lymphocytes were collected. . Results The expression of mTOR and raptor in cancer tissues was significantly higher than that in paracancerous tissues. The positive rate of clinical stage Ⅲ ~ Ⅳ mTOR protein was significantly higher than that of clinical stage Ⅰ ~ Ⅱ. The expression of p-mTOR and p-TSC2 in peripheral blood T lymphocytes of patients was higher than that of normal people. Conclusion The activation of mTOR signaling pathway in esophageal squamous cell carcinoma and peripheral T lymphocytes is stronger than normal.
