目的探讨反复自然流产患者辅助性T细胞17(Th17)免疫应答情况及其与Th1、Th2和Treg细胞相互之间的关系。方法实时荧光定量PCR(Realtime PCR)检测反复自然流产患者30例(观察组)及健康早孕体检者38例(对照组)外周血中的IL-17、T-bet、GATA-3和FoxP3 mRNA表达水平。结果观察组IL-17mRNA、T-bet mRNA表达水平较对照组明显升高,差异有统计学意义(P<0.0001)。而GATA-3 mRNA、FoxP3 mRNA表达水平显著低于对照组(P<0.0001);观察组Th1/Th2和Th17/Treg的比值均较对照组显著升高,差异有统计学意义(P<0.0001)。结论 Th17细胞参与了反复自然流产的免疫应答,Th1/Th2和Th17/Treg细胞之间的平衡在妊娠的维持中发挥重要作用。
Objective To investigate the immune response of helper T cells (Th17) in patients with recurrent spontaneous abortion and its relationship with Th1, Th2 and Treg cells. Methods Real-time PCR was used to detect the mRNA expression of IL-17, T-bet, GATA-3 and FoxP3 in peripheral blood of 30 patients with spontaneous abortion (observation group) and 38 healthy controls (control group) Level. Results The expression levels of IL-17 mRNA and T-bet mRNA in the observation group were significantly higher than those in the control group (P <0.0001). (P <0.0001). The ratios of Th1 / Th2 / Th17 / Treg in the observation group were significantly higher than those in the control group (P <0.0001), while the levels of GATA-3 mRNA and FoxP3 mRNA in the observation group were significantly lower than those in the control group . Conclusion Th17 cells are involved in the immune response to recurrent spontaneous abortion. The balance between Th1 / Th2 and Th17 / Treg cells plays an important role in the maintenance of pregnancy.
