目的:探讨茎叶人参皂甙对内毒素所致的急性肺损伤大鼠血小板活化因子(PAF)水平的影响。方法:SD大鼠30只,随机分为正常组、内毒素组、人参皂甙组各10只,实验开始后内毒素组和人参皂甙组均尾静脉注射内毒素脂多糖,给药5小时后,经腹主动脉取血行血气分析,然后处死大鼠,肉眼观察肺组织改变,计算肺系数、肺湿/干重比(W/D)及病理学检查,ELISA法测肺组织匀浆中PAF水平。结果:与正常组比较,内毒素组大鼠一般状况较差,肉眼观察肺组织水肿出血明显,肺W/D升高,动脉血氧分压(PaO_2)显著下降,病理学检查肺组织明显损伤,PAF水平明显升高;人参皂甙组一般状况较好,肉眼观察肺组织水肿出血较轻,病理学检查肺组织损伤较轻,其PaO_2、PaO2/FiO_2、肺W/D、肺系数值及肺组织中PAF、BALF水平较内毒素组明显改善(P<0.05)。结论:茎叶人参皂甙对急性肺损伤的保护作用机制可能与其抑制PAF水平有关。
Objective: To investigate the effects of ginsenosides in stems and leaves on platelet activating factor (PAF) levels in rats with acute lung injury induced by endotoxin. METHODS: Thirty SD rats were randomly divided into normal group, endotoxin group, and ginsenoside group. Ten rats were injected with endotoxin lipopolysaccharide in the endotoxin group and ginsenoside group after 5 hours. Blood samples were taken from the abdominal aorta for blood gas analysis. The rats were then sacrificed and their lung tissue changes were visually observed. Lung coefficient, lung wet/dry weight ratio (W/D) and pathological examinations were calculated. PAF levels in lung homogenates were measured by ELISA. . RESULTS: Compared with the normal group, the rats in the endotoxin group had poor general conditions. The edema in the lung tissue was observed with the naked eye, the W/D in the lung was increased, and the PaO 2 in the arterial blood was significantly decreased. The lung tissue was significantly damaged by pathological examination. The PAF level was significantly elevated; the ginsenoside group was generally in good condition, the lungs were observed for edema and bleeding were minor, and the pathological examination for the lung tissue was lighter, with PaO2, PaO2/FiO2, lung W/D, lung coefficient values, and lungs. The levels of PAF and BALF in the tissue were significantly improved compared with the endotoxin group (P<0.05). Conclusion: The protective mechanism of stem and leaf ginsenosides on acute lung injury may be related to its inhibition of PAF levels.
