目的寻找骨肉瘤转移相关基因,初步分析Matrilin-3基因表达与骨肉瘤转移的关系。方法通过RT-PCR、Westen-blot及免疫组化检测该基因在不同骨肉瘤细胞系、正常成骨细胞、骨肉瘤及正常骨组织临床标本中的表达,并分析其与骨肉瘤患者预后的关系。结果 Matrilin-3在高转移MG36-A1细胞系中表达明显下降。RT-PCR、Westen-blot检测发现Matrilin-3在正常成骨细胞中高表达,在SAOS2、143B中表达缺失。免疫组化检测显示在25例骨肉瘤临床标本中,24%(6/25例)的标本Matrilin-3表达为阳性,其余为阴性;在正常骨组织中Matrilin-3表达强阳性。Matrilin-3表达与骨肉瘤患者预后呈正相关(P=0.023)。结论 Matrilin-3的表达随着骨肉瘤细胞系转移能力的增加而出现缺失,Matrilin-3可能成为阻遏骨肉瘤转移的新靶点。
OBJECTIVE: To search for the genes related to metastasis of osteosarcoma and to analyze the relationship between Matrilin-3 gene expression and metastasis of osteosarcoma. Methods RT-PCR, Westen-blot and immunohistochemistry were used to detect the expression of this gene in clinical specimens of different osteosarcoma cell lines, normal osteoblasts, osteosarcoma and normal bone tissues, and to analyze their relationship with the prognosis of patients with osteosarcoma . Results Matrilin-3 was significantly down-regulated in the highly metastatic MG36-A1 cell line. RT-PCR and Westen-blot showed that Matrilin-3 was overexpressed in normal osteoblasts and was absent in SAOS2 and 143B. Immunohistochemistry showed that Matrilin-3 was positive in 24% (6/25) of the 25 specimens of osteosarcoma, and negative in the other specimens. Matrilin-3 was strongly positive in normal bone tissues. Matrilin-3 expression was positively correlated with the prognosis of patients with osteosarcoma (P = 0.023). Conclusions The expression of Matrilin-3 is absent in osteosarcoma cell lines. Matrilin-3 may be a new target for the suppression of osteosarcoma metastasis.
