对聚乙二醇诱导的细胞融合机制进行了初步探讨,认为:在细胞融合中,聚乙二醇的作用不仅表现在其“亲水性”上,更重要的是其“亲脂性”。聚乙二醇分子的亲水,亲脂两性性质在细胞融合的膜变化中,起着决定性作用。本文还以聚乙二醇二羧酸为母体,合成了四种聚乙二醇脂类衍生物,并以艾氏腹水瘤为材料测定了其融合活性。其中化合物聚乙二醇6000二羧酸—胆固醇酯的融合指数是聚乙二醇6000的2.7倍、聚乙醇6000二羧酸—葡萄糖是聚乙二醇的4.3倍。
The mechanism of cell fusion induced by PEG was preliminarily discussed. It is believed that the role of PEG in cell fusion is not only “hydrophilic” but more importantly, it is “lipophilic”. Hydrophilic, lipophilic amphipathic properties of polyethylene glycol molecules play a decisive role in membrane changes in cell fusion. In this paper, polyethylene glycol dicarboxylic acid as a parent, four kinds of polyethylene glycol lipid derivatives were synthesized, and the Ehrlich ascites tumor activity was measured. The compound polyethylene glycol 6000 dicarboxylic acid - cholesterol ester fusion index is 2.7 times that of polyethylene glycol 6000, polyethylene glycol 6000 dicarboxylic acid - glucose is 4.3 times that of polyethylene glycol.
