目的 :研究紫草素对人胎肾小球系膜细胞(MC)增殖、凋亡及细胞外基质的影响。方法 :采用人胎肾细胞培养法、MTT法、流式细胞仪及免疫组化的方法。结果 :紫草素 0 .0 5、0 .10、0 .5 0及 1.0 0mmol L浓度对培养 2 4、4 8、72h人MC有明显抑制作用 ;紫草素明显抑制高糖诱发的人MC细胞凋亡 ;紫草素 0 .0 5、0 .10、0 .5 0 μg ml显著抑制系膜基质层粘连蛋白 (LN)、纤维连接蛋白 (FN)及胶原Ⅳ (CoⅣ )的生成。结论 :紫草素具抑制人MC增殖、细胞外基质生成及细胞凋亡作用。
Objective : To study the effects of shikonin on proliferation, apoptosis and extracellular matrix of human fetal mesangial cells (MC). Methods: Human fetal kidney cell culture method, MTT assay, flow cytometry and immunohistochemistry were used. Results: Shikonin concentrations of 0.05, 0.10, 0.50, and 1.0 mmol L significantly inhibited MC in 24, 48, and 72 h cultures; shikonin significantly inhibited MC induced by high glucose. Apoptosis; shikonin 0.05, 0.10, 0.50 μg ml significantly inhibited the formation of mesangial laminin (LN), fibronectin (FN) and collagen IV (CoIV). Conclusion: Shikonin can inhibit the proliferation of human MC, the production of extracellular matrix and apoptosis.
