目的 :观察角质细胞生长因子 (KGF)在体外对常染色体显性遗传型多囊肾病 (ADPKD)囊肿衬里上皮细胞的促增殖作用并探讨其可能的作用机制。方法 :外源性给予不同浓度的 KGF(0~ 10 0 ng/ ml) ,采用 MTT法检测细胞增殖情况 ,流式细胞术检测细胞周期 ,在电镜下观察细胞形态。结果 :KGF呈剂量、时间依赖性地促进了 ADPKD囊肿衬里上皮细胞的增生 ,表现为细胞数量增多 ,S期细胞比率增高 ,细胞分裂活跃 ,增生的细胞形态尤其是细胞核较正常体积增大。结论 :KGF可显著刺激 ADPKD囊肿衬里上皮细胞的增殖 ,提示 KGF可能参与 ADPKD肾囊肿的形成和发展。
OBJECTIVE: To observe the effect of KGF on the proliferation of cyst-lining epithelial cells in autosomal dominant polycystic kidney disease (ADPKD) in vitro and to explore its possible mechanism. Methods: Exogenous KGF (0 ~ 10 0 ng / ml) was administered exogenously. Cell proliferation was detected by MTT assay. Cell cycle was detected by flow cytometry. Cell morphology was observed under electron microscope. Results: KGF promoted the proliferation of epithelial cells in the lining of ADPKD cysts in a time-and dose-dependent manner. The number of cells increased, the proportion of S phase cells increased, the cell division was active, and the cell morphology, especially the nucleus, increased compared with normal volume. Conclusion: KGF can significantly stimulate the proliferation of epithelial cells in the lining of ADPKD cysts, suggesting that KGF may be involved in the formation and development of renal cysts in ADPKD.
