高尔基法已有100余年的历史,但至今其染色结果仍不很稳定,较难掌握。由于高尔基法在显示完整的神经元和神经元间联系上具有独特的优点,目前常被采用。所以近年来,改进之法不断增加,而且有应用于电镜观察的方法问世。高尔基法显示细胞形态和细胞间联系存在问题之一,就是大块材料的浸染不能得到满意的结果。但大块浸染对研究中枢神经系统又是不可缺少的。近年来,我们在浸染大块神经组织方面进行了改进,获得满意的结果,现将我们的经验和体会总结如下。我们常做的材料的大小为厚度5-6mm,长和宽皆为30-35 mm。1.动物-常用10-20天小兔,7-9天小白鼠及成年猫。2.固定方法-用磷酸缓冲液配制1%多聚甲醛,1.25%戊二醛为固定液或用0.25%戊二醛固定液,进行灌流固定,两种固定液均可得到较好的效果。判定灌流
