将楮头红醇提取物用正丁醇萃取,萃取物经硅胶柱层析和制备型高效液相色谱(PHPLC)纯化得一纯净物,通过IR、ESI-MS和13C NMR谱等分析,鉴定为呋甾烷型甾体皂苷,命名为楮头红皂苷A(SWSA),系首次从楮头红中提取得到.抑菌试验结果表明:SWSA对金黄色葡萄球菌、枯草芽孢杆菌、耶尔森氏菌和嗜水气单胞菌均有较好的抑制效果,其最小抑菌浓度分别为2.8、3.3、2.5和2.2 mg·m L-1;对大肠埃希氏菌和铜绿假单胞菌的抑制效果不明显.通过3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)试验检测SWSA对Hep G2.2.15细胞的毒性,采用酶联免疫吸附试验(ELISA)法测定各组细胞培养上清液中HBs Ag和HBe Ag的水平,综合评价不同含量SWSA体外抗乙肝病毒(HBV)的活性.结果表明,SWSA处理Hep G2.2.15细胞后,HBs Ag和HBe Ag的表达均呈时间和剂量依赖性抑制,治疗指数分别为6.43和4.23,预示SWSA具有一定的体外抗HBV活性.
The extract of Quercus rubra was extracted with n-butanol. The extract was purified by silica gel column chromatography and preparative high performance liquid chromatography (PHPLC).
