本试验选用不同类型细胞缺陷小鼠,将其分成六个试验组,前三组为CBA/N与BALB/c小鼠交配所生仔一代,是B细胞免疫功能缺陷及其相应的对照小鼠,后三组为615B6/PB1-nu与C57B1/6-bg交配所生之仔鼠,是T淋巴细胞免疫功能缺陷与T和NK细胞联合免疫功能缺陷小鼠及其对照小鼠,以上各组小鼠分别以1亿菌体布氏活菌苗注射小鼠腹腔,于注射后一定间隔检测抗体效价并取有关脏器做细菌培养,B细胞功能缺陷小鼠抗体效价为1:80-1:160,对照小鼠为1:160-1:640,T细胞和T、NK细胞功能缺陷小鼠抗体效价为1:4-1:80。对照小鼠为1:128-1:640,由于布氏菌是细胞内增殖,细菌主要存在于脾脏,故于45天时心血培养虽为阴性而脾脏仍有细菌生长,脾重的增减与抗体效价有一定关系,与活菌数不成平行关系,该小鼠为菌苗免疫机理探讨提供了模型。 CBA/N小鼠为X-链隐性突变,基因符号为Xid(X-Linked immunodeficient),此隐性突变基因位于X-性染色体上,免疫调节基因也在此染色体上,该基因的隐性突变直接影响免疫细胞和免疫球蛋白的产生。CBCF_1是CBA/N与BALB/c鼠交配所生之仔一代,其基因型雌性为Xid/x是免疫功能正常小鼠,雄性为Xid/y具有B细胞功能缺陷特性,可用于研究B细胞的发生,功能和异质性理想的动物模型。 615B6/PB1-nu为T淋巴细胞功能缺陷型小鼠(简称裸鼠),61586/PBt-bg/nu为T和NK细胞联合免疫功能缺陷型小鼠(简称beige裸鼠),该裸鼠和beige裸鼠是615/PB1-+/bg·+/nu♀与C_(57)BI/6-bg/bg·nu/nu♂交配所生之仔鼠,615/PB1近交系小鼠是经无菌剖腹产所得之仔鼠。上述小鼠用布氏活菌苗免疫,对其免疫后的反应进行观察,为以上小鼠的实验应用提供参考依据。
