为探讨云芝多糖对巨噬细胞一氧化氮产生的影响,采用Gries试剂法对小鼠腹腔巨噬细胞培养上清液中一氧化氮的含量进行测定,并用结晶紫染核法测定细胞数目。结果发现,在不受任何刺激的情况下,云芝多糖活化的巨噬细胞一氧化氮的释放并不增加;而在脂多糖作用下,其一氧化氮产量明显增加;云芝多糖活化的巨噬细胞与γ干扰素共孵,并未见一氧化氮产量增加。提示云芝多糖对巨噬细胞的激活采用的方式与γ干扰素类似,其抗动脉粥样硬化的效应可能部分源自于其对巨噬细胞一氧化氮生成的调节。
In order to investigate the effect of polysaccharide of Coriolus versicolor on nitric oxide production in macrophages, the content of nitric oxide in peritoneal macrophages culture supernatant was determined by Gries reagent, and the number of cells was determined by crystal violet staining. The results showed that without any stimulation, PSK-activated macrophages did not increase the release of nitric oxide, while the production of nitric oxide increased significantly under the action of lipopolysaccharide; Phagocytic cells and γ interferon incubation, and no increase in nitric oxide production. Tip Coriolus versicolor polysaccharide activation of macrophages and interferon-γ in a similar manner, its anti-atherosclerotic effect may be due in part to its regulation of macrophage nitric oxide production.
