目的研究丹参酮Ⅱ-A对大鼠心肌细胞L-型钙通道的影响。方法采用酶解法制备大鼠心肌细胞,应用全细胞膜片钳技术检测大鼠心肌细胞L-型钙通道的电流密度和动力学。结果丹参酮Ⅱ-A对L-型钙通道有抑制作用。在0μmol/L、10μmol/L、20μmol/L和40μmol/L丹参酮Ⅱ-A作下,L-型钙通道的峰值电流密度分别为-13.34±1.06pA/pF、-10.46±0.75pA/pF、-7.62±0.50PA/pF和-4.69±0.18PA/pF,其抑制率分别为28.40%、52.14%和73.50%(n=8,P<0.05)。结论丹参酮Ⅱ-A抑制大鼠心肌细胞L-型钙通道,降低L-型钙通道的电流密度,呈浓度依赖性。
Objective To investigate the effect of tanshinone Ⅱ-A on L-type calcium channel in rat cardiomyocytes. Methods Rat cardiomyocytes were prepared by enzymolysis. Whole-cell patch-clamp technique was used to detect the current density and kinetics of L-type calcium channel in rat cardiomyocytes. Results Tanshinone Ⅱ-A inhibited L-type calcium channel. The peak current densities of L-type calcium channels were -13.34 ± 1.06pA / pF and -10.46 ± 0.75pA / pF at 0μmol / L, 10μmol / L, 20μmol / L and 40μmol / -7.62 ± 0.50PA / pF and -4.69 ± 0.18PA / pF respectively. The inhibition rates were 28.40%, 52.14% and 73.50%, respectively (n = 8, P <0.05). Conclusion Tanshinone Ⅱ-A can inhibit the L-type calcium channel of rat myocardial cells and decrease the current density of L-type calcium channel in a concentration-dependent manner.
