目的探讨TRβ1在人类乳腺癌组织及癌旁组织的表达情况及其在乳腺癌组织中表达的临床意义。方法收集112例新疆医科大学第五附属医院和附属肿瘤医院经临床证实的乳腺癌患者的乳腺癌组织及癌旁组织石蜡标本,使用免疫组织化学链霉素抗生物素-过氧化物酶(SP)法检测112例乳腺癌组织及癌旁组织中TRβ1蛋白的表达,并调取112例乳腺癌患者临床资料,分析乳腺癌组织中TRβ1蛋白表达与临床病理资料之间的关系。结果 TRβ1阳性表达主要定位于细胞核和细胞质。TRβ1在人类乳腺癌组织中阳性表达显著低于癌旁正常组织,乳腺癌组织中TRβ1蛋白阳性表达率为47.32%(53/112),显著低于癌旁正常组织(100%,112/112);乳腺癌组织中TRβ1蛋白异常低表达与淋巴结转移(χ~2=14.837,P<0.001)、组织学分级(χ~2=8.459,P=0.015)及Her-2状态(χ~2=4.579,P=0.032)、临床分期(χ~2=4.177,P=0.041)密切相关,与年龄、民族、原发肿瘤大小、ER、PR、基因分型无关(P>0.05)。结论相比癌旁组织,乳腺癌组织中TRβ1阳性表达低;乳腺癌组织学分级高者TRβ1阳性表达率低;TRβ1阴性表达与临床分期晚、淋巴结转移等乳腺癌不良预后因素相关;在乳腺癌组织TRβ1阳性表达与乳腺癌分子分型及预后无关。
Objective To investigate the expression of TRβ1 in human breast cancer tissues and adjacent tissues and its clinical significance in breast cancer tissues. Methods Paraffin-embedded specimens of breast cancer tissues and paracancerous tissues were collected from 112 cases of clinically proven breast cancer patients in the fifth affiliated hospital of Affiliated Hospital of Xinjiang Medical University and its affiliated cancer hospital. Immunohistochemical streptavidin-peroxidase (SP) was used. The expression of TRβ1 protein was detected in 112 breast cancer tissues and paracancerous tissues, and the clinical data of 112 breast cancer patients were detected. The relationship between the expression of TRβ1 protein and clinicopathological data was analyzed. Results The positive expression of TRβ1 was mainly located in the nucleus and cytoplasm. The positive expression of TRβ1 in human breast cancer tissues was significantly lower than that in adjacent normal tissues. The positive rate of TRβ1 protein in breast cancer tissues was 47.32% (53/112), which was significantly lower than that in normal tissues (100%, 112/112). The abnormal expression of TRβ1 protein in breast cancer tissues was correlated with lymph node metastasis (χ~2=14.837, P<0.001), histological grade (χ~2=8.459, P=0.015) and Her-2 status (χ~2=4.579). P=0.032) and clinical stage (χ~2=4.177, P=0.041) were closely related to age, nationality, primary tumor size, ER, PR and genotyping (P>0.05). Conclusions The positive expression of TRβ1 in breast cancer tissues is lower than that in adjacent tissues, and the positive expression rate of TRβ1 is lower in breast cancers with higher histological grades. The negative expression of TRβ1 is associated with poor prognostic factors in late stage breast cancer and lymph node metastasis. The positive expression of TRβ1 in tissues was not associated with breast cancer molecular typing and prognosis.
