目的:研究锌指蛋白(ZNF)185在小鼠不同发育阶段睾丸中的表达。方法:从小鼠脑、肝、卵巢和不同发育阶段睾丸组织中提取总RNA,采用半定量RT-PCR法检测ZNF185的表达;制备小鼠不同发育阶段睾丸冰冻切片,应用免疫细胞化学分析法检测ZNF185蛋白的定位。结果:①ZNF185在睾丸中的表达显著高于脑、肝和卵巢组织。②ZNF185在小鼠性成熟前组睾丸中的表达显著低于性成熟期和老年期,而性成熟期和老年期之间差异不显著。③在性成熟前,ZNF185主要定位于睾丸间质细胞和类肌细胞;在性成熟期和老年期小鼠中,ZNF185主要定位于睾丸间质细胞、类肌细胞和精子。结论:ZNF185在小鼠性成熟后睾丸中的表达升高,定位更广泛。
Objective: To study the expression of zinc finger protein (ZNF) 185 in testes at different developmental stages of mice. Methods: Total RNA was extracted from the testis of brain, liver, ovary and different developmental stages in mice. The expression of ZNF185 was detected by semi-quantitative RT-PCR. Frozen sections of testes were prepared at different developmental stages. Immunocytochemistry was used to detect the expression of ZNF185 Protein localization. Results: ① The expression of ZNF185 in testis was significantly higher than that in brain, liver and ovary. ② The expression of ZNF185 in testes of mouse premature mature group was significantly lower than that in sexual maturity and old age, while there was no significant difference between sexual maturity and old age. ③ Before sexual maturation, ZNF185 mainly located in testicular stromal cells and myoid cells; in sexual maturity and old mice, ZNF185 mainly located in testicular stromal cells, myoid cells and sperm. Conclusion: The expression of ZNF185 in testes after sexual maturation in mice is increased and its localization is more extensive.
