目的:探讨叶酸缺乏对胚胎大鼠神经干细胞(NSCs)蛋白质表达的影响。方法:采用无血清体外细胞培养方法,培养胚胎大鼠NSCs并进行鉴定,采用叶酸缺乏培养基培养NSCs,根据叶酸添加终浓度将细胞分为对照(Con?trol)组、叶酸缺乏(Folate-D)组、叶酸补充(Folate-L)组,各组叶酸终浓度分别为4、0.6、8mg/L。应用双向电泳(2-DE)技术分析各组蛋白质表达的差异。结果:建立了稳定可重复的NSCs2-DE图谱,与Control组相比,Folate-D组2个蛋白点表达量降低,4个蛋白点表达量升高;Folate-L组2个蛋白点表达量降低,1个蛋白点表达量升高。结论:叶酸缺乏或补充叶酸影响NSCs蛋白表达,差异蛋白分离为进一步研究叶酸对NSCs增殖分化调控机制奠定了基础。
Objective: To investigate the effect of folic acid deficiency on protein expression of neural stem cells (NSCs) in embryonic rat. Methods: NSCs were cultured and cultured in serum-free culture in vitro. NSCs were cultured in folic acid-deficient medium. The cells were divided into control group (Con trol group) and Folate-D ) Group and Folate-L group. The final concentrations of folate in each group were 4,0.6 and 8 mg / L, respectively. Two-dimensional electrophoresis (2-DE) technique was used to analyze the difference of protein expression in each group. Results: A stable and reproducible NSCs2-DE map was established. Compared with Control group, the expression of two protein spots in Folate-D group decreased and the expression of four protein spots increased. The expression of two protein spots in Folate-L group Reduce, a protein expression increased point. CONCLUSION: Folic acid deficiency or folic acid affects the expression of NSCs protein, and the differential protein separation may provide a basis for further study on the regulatory mechanism of folic acid on the proliferation and differentiation of NSCs.
