目的从新生大鼠大脑皮质中分离、培养并鉴定星形胶质细胞。方法分离出生1 ̄3d的大鼠大脑皮质细胞加入含有10%马血清、10%小牛血清以及10ng/mLEGF的MEM培养基培养,于第3 ̄4周将细胞固定后采用免疫细胞化学技术检测特异性星形胶质细胞抗原GFAP的表达。结果从新生大鼠大脑皮质分离的细胞生长旺盛,星形胶质细胞形态典型,并表达星形胶质细胞特异性抗原GFAP,未见其他种类细胞生长。结论笔者建立了一种简单、可靠的大鼠星形胶质细胞的体外培养方法,为应用胶质细胞饲养层与其他细胞联合培养奠定了基础。
Objective To isolate, culture and identify astrocytes from neonatal rat cerebral cortex. Methods Rat cerebral cortical cells from 1 to 3 days of age were isolated and cultured in MEM medium containing 10% horse serum, 10% calf serum and 10 ng / mL EGF. After 3 to 4 weeks, cells were fixed and detected by immunocytochemistry Specific astrocyte antigen GFAP expression. Results The cells isolated from cerebral cortex of neonatal rats grew vigorously, with typical morphology of astrocytes and expressed astrocyte-specific antigen GFAP. No other kind of cell growth was observed. Conclusion The author established a simple and reliable in vitro culture method of rat astrocytes, laying the foundation for the co-culture of glial cell feeder layer and other cells.
