目的:建立中药炮制辅料猪胆汁中胆红素含量测定方法。方法:采用Zorbax RX-C18色谱柱(4.6mm×250mm,5μm);以二甲基亚砜-乙腈-0.125mol·L-1醋酸铵水溶液(60∶60∶70)为流动相,其中醋酸铵溶液用冰醋酸调pH=5.3;流速1.0mL·min-1;检测波长452nm。结果:胆红素进样量在0.0113~0.1356μg范围内呈良好线性关系(r=0.9998);平均回收率(n=6)为98.13%,RSD为2.5%。结论:本方法准确、简便,重复性好,可为中药炮制辅料猪胆汁的质量控制提供科学依据。
Objective: To establish a method for the determination of bilirubin in pig bile. Methods: A Zorbax RX-C18 column (4.6mm×250mm, 5μm) was used. The mobile phase consisted of dimethylsulfoxide-acetonitrile-0.125mol·L-1 ammonium acetate solution (60∶60∶70). Ammonium acetate was used as mobile phase. The solution was adjusted to pH=5.3 with glacial acetic acid; the flow rate was 1.0 mL·min-1; and the detection wavelength was 452 nm. Results: There was a good linear relationship (r=0.9998) in the range of 0.0113-0.1356 μg for bilirubin injection. The average recovery (n=6) was 98.13% and the RSD was 2.5%. Conclusion: The method is accurate, simple, and reproducible, and can provide scientific basis for the quality control of pig bile made from Chinese herbal medicine.
