目的 研究辐射敏感小鼠乳腺癌细胞SX 9的ku70、ku80基因型 ,初步探讨ku基因突变对其辐射敏感性的影响。方法 RT PCR克隆并鉴定ku70 /ku80全长基因 ,并对其全长进行全自动测序。脂质体转染正常人全长ku80cDNA至SX 9细胞 ,克隆形成比较转染了正常ku80基因的SX 9与SX 9以及SR 1细胞存活率。结果 SX 9细胞ku70基因正常 ,而其ku80存在基因突变 ,转染了正常人全长ku80cDNA的SX 9细胞能部分重建其对γ射线的存活率。结论 SX 9细胞ku基因发生突变 ,这可能是其DNA损伤修复缺陷并导致对辐射敏感的分子基础
Objective To study the ku70 and ku80 genotypes of SX9 in radiation-sensitive mouse breast cancer cells, and to explore the effect of Ku mutation on its radiation sensitivity. Methods The full-length ku70 /ku80 gene was cloned and identified by RT PCR and its full length was fully sequenced. The human full-length ku80 cDNA was transfected into SX9 cells with liposomes, and the survival rate of SX9 and SX9 cells and SR1 cells transfected with the normal ku80 gene were compared. Results The ku70 gene was normal in SX9 cells, and there was a mutation in ku80 gene. SX9 cells transfected with full-length human ku80 cDNA could partially reconstruct their survival rate for γ-rays. Conclusion The mutation of ku gene in SX9 cells may be the molecular basis of its defects in DNA damage repair and lead to radiation sensitivity.
