研究视交叉上核内主要肽能神经元的分布情况,选用健康成年雄性SD大鼠5只,以免疫组化双重及三重标记技术,对大鼠视交叉上核(SCN)内血管活性肠多肽(VIP)、精氨酸血管加压素(AVP)及生长抑素(SOM)样三类神经细胞群的分布进行了观察。免疫染色以ABC法进行,呈色底物采用DAB、TMB及4-氯-1-萘酚。结果显示:在SCN头中尾三段,三种肽能神经元的数量及分布均存在差异;VIP与AVP样细胞群之间并不是截然分界,其相交处一狭窄区内三种肽能神经元混杂在一起,交错重叠。上述发现为SCN机能学的研究提供了新的形态学依据。
To study the distribution of major peptide neurons in the suprachiasmatic nucleus, five healthy adult male Sprague-Dawley rats were used to detect the expression of vasoactive intestinal polypeptide (PCNA) in the suprachiasmatic nucleus (SCN) by immunohistochemical double and triple labeling (VIP), arginine vasopressin (AVP) and somatostatin (SOM) -like three kinds of neuronal cell population were observed. Immunostaining with ABC method, the color of the substrate using DAB, TMB and 4-chloro-1-naphthol. The results showed that there were differences in the number and distribution of the three kinds of peptidergic neurons in the third tail of the SCN head. The VIP and AVP-like cell populations were not completely demarcated. Three kinds of peptidergic neurons Mixed together, staggered overlap. The above findings provide a new morphological basis for the study of SCN functional properties.
