目的:观察免疫刺激DNA序列(ISS)与尘螨过敏原(Df)合用,对Df致敏的变应性鼻炎(AR)大鼠模型鼻腔过敏性炎症的作用及其维持时间。方法:SD大鼠32只,分ISS组(A组)、ISS加Df组(B组)、Df组(C组)和正常对照组(D组),每组各8只。A、B、C组用Df致敏及激发。各组在第1次激发后6周中,每周采血1次测特异性IgE,最后处死大鼠,组织病理学方法观察鼻黏膜改变,ELISA测血清IL-4、IL-5、IFN-γ和IL-12水平。结果:A、B组IFNγ-和IL-12水平明显高于C组(P<0.01),而A、B组IL-4和IL-5水平明显低于C组(P<0.01)。血清特异性IgE前5周B组与C组比较差异有统计学意义(P<0.01),第6周B组Df特异性IgE水平有回升,但与C组比较差异仍有统计学意义(P<0.05);而A组与C组比较差异无统计学意义(P>0.05)。结论:ISS与Df合用对Df致敏的AR大鼠模型鼻腔过敏性炎症有很好的抑制作用,其抑制作用可维持6周。
OBJECTIVE: To observe the effect of immunostimulatory DNA (ISS) combined with dust mite allergen (Df) on nasal allergic inflammation in Df-sensitized allergic rhinitis (AR) rat model and its maintenance time. Methods: Thirty-two SD rats were divided into three groups: ISS group (A group), ISS plus Df group (B group), Df group (C group) and normal control group (D group). A, B, C group with Df sensitization and excitation. The specific IgE was collected once a week for 6 weeks after the first challenge in each group, and finally the rats were sacrificed. The changes of nasal mucosa were observed by histopathology. The levels of IL-4, IL-5, IFN-γ And IL-12 levels. Results: The levels of IFNγ- and IL-12 in group A and group B were significantly higher than those in group C (P <0.01), while the levels of IL-4 and IL-5 in group A and group B were significantly lower than those in group C (P <0.01). Serum-specific IgE in the first 5 weeks of group B compared with the C group was statistically significant (P <0.01), the sixth week of B group Df-specific IgE levels rise, but compared with the C group, the difference was statistically significant (P <0.05). There was no significant difference between group A and group C (P> 0.05). Conclusion: The combination of ISS and Df can inhibit nasal allergic inflammation in Df-sensitized AR rat model, and its inhibitory effect can be maintained for 6 weeks.
