为获得微拟球藻不同时期转录组的信息,揭示基因表达差异与油脂积累的关系。采用Illumina HiSeq 2000高通量测序技术,转录组在指数期和平台期对微拟球藻进行测序分析,拼接组装后共获得29 203条序列,指数期表达上调9745条,平台期表达上调7 305条。通过NCBI数据库进行Blast同源性比对,有13 775条序列能获得同源序列注释。通过KEGG pathway数据库比对,指数期涉及238种代谢途径,表达上调的序列有818条;平台期涉及248种代谢途径,表达上调的序列有1 148条;两时期关于脂质代谢的序列共有195条,指数期表达上调53条,平台期表达上调47条。通过COG数据库比对,将具有COG功能注释的11 604条序列分为25大类,涉及物质及信号传导、能量产生与转化、氨基酸代谢等诸多生理生化过程。
In order to obtain the information of transcriptome of different stages of Nannochloropsis, the relationship between gene expression and lipid accumulation was revealed. Using Illumina HiSeq 2000 high-throughput sequencing, the transcriptome was sequenced in the exponential phase and plateau stage. 29 203 sequences were obtained after splicing and assembly, with 9,745 up-regulated in exponential phase and 7 305 up-regulated in plateau phase Article. Blast homology alignment was carried out through the NCBI database, and 13 775 sequences could be annotated with homologous sequences. Through the KEGG pathway database comparison, 238 metabolic pathways were involved in the exponential phase, with 818 up-regulated sequences. There were 248 metabolic pathways involved in the plateau phase, with 1 148 up-regulated sequences in the plateau phase. There were 195 sequences involved in lipid metabolism during the two phases Article, the exponent expresses up 53, platform stage expresses up 47. Through the comparison of COG database, 11 604 sequences with COG functional annotation were divided into 25 categories, involving many physiological and biochemical processes such as substance and signal transduction, energy production and transformation, amino acid metabolism and so on.
