采用免疫组化及原位分子杂交技术,本文研究了负重力竭性游泳对大鼠胰岛A、B细胞分泌功能的影响。主要结果发现:力竭状态下,①大鼠胰岛Ins-IR细胞呈色反应强度增加,而Glu-IR细胞呈色反应强度降低,说明B细胞Ins释放被抑制,A细胞Glu释放增多。②大鼠胰岛B细胞内Pro-InsmRNA杂交信号明显减弱,说明Pro-Ins-mRNA转录受到抑制,Ins合成受抑。③Ins-IR和IAPP-IR细胞未呈现相同程度的呈色反应变化,提示B细胞内存在着含IAPP/Ins不同比率的分泌颗粒,可随机体的机能状态需要而分别释放。
Using immunohistochemistry and in situ hybridization, we investigated the effect of exhaustive swimming on the secretion of rat pancreatic islet A and B cells. The main findings were as follows: ① In the exhaustive state, ① the intensity of the color reaction of Ins-IR cells in rat islets increases, while the intensity of the color reaction in Glu-IR cells is decreased, which indicates that Ins release from B cells is inhibited and Glu release from A cells is increased. ② Pro-Ins mRNA hybridization signal in rat pancreatic islet B cells was significantly weakened, indicating that Pro-Ins-mRNA transcription was inhibited and Ins synthesis was inhibited. ③Ins-IR and IAPP-IR cells did not show the same degree of color reaction changes, suggesting that there are secreted particles with different ratios of IAPP / Ins in B cells, which may be released when the functional status of the cells needs to be changed.
