目的:探讨舒郁宁心汤抗抑郁功效的作用机理。方法:40只SD大鼠随机分为正常组、模型组、中药组、氟西汀组。建立慢性应激抑郁大鼠模型。TUNEL法检测大鼠海马凋亡细胞,免疫组化方法观察大鼠海马Bcl-2表达的变化。结果:模型组海马CA3区细胞凋亡数比正常组增加,差异有统计学意义(P<0.01);舒郁宁心中药组、氟西汀组海马CA3区细胞凋亡数较模型组减少,差异有统计学意义(P<0.01)。与空白对照组比较,模型组大鼠海马Bcl-2表达降低,差异有统计学意义(P<0.01或P<0.05);与模型组相比中药组、氟西汀组大鼠海马区Bcl-2表达升高,差异有统计学意义(P<0.01或P<0.05)。结论:舒郁宁心汤可能通过减少海马神经元细胞凋亡,发挥抗抑郁作用。
Objective: To investigate the mechanism of antidepressant effect of Shuyu Ningxin Decoction. Methods: Forty SD rats were randomly divided into normal group, model group, TCM group and fluoxetine group. To establish a rat model of chronic stress depression. The apoptosis of hippocampus was detected by TUNEL method. The expression of Bcl-2 in hippocampus was observed by immunohistochemistry. Results: The number of apoptotic cells in hippocampal CA3 area of model group was higher than that of normal group (P <0.01), and the number of apoptotic cells in hippocampal CA3 area of Shuyu Ningxin Herbal Medicine group and Fluoxetine group was lower than that of model group Statistical significance (P <0.01). Compared with the blank control group, the expression of Bcl-2 in the hippocampus in the model group decreased significantly (P <0.01 or P <0.05). Compared with the model group, Bcl- 2 expression increased, the difference was statistically significant (P <0.01 or P <0.05). Conclusion: Shu Yu Ning Xin Tang may play an antidepressant effect by reducing apoptosis in hippocampal neurons.
