本实验将急性非淋巴细胞白血病病人的骨髓细胞进行扩散盒培养。将受鼠分为对照组及用药组,对用药组及对照组行光镜观察,计数集落。将对照组及用药组中含有集落的琼脂块取出做电镜观察。结果表明,正常骨髓的粒单系祖细胞(CFU-GM)集落由成熟细胞组成:白血病骨髓的L-CFU集落具有明显的白血病性质;氨甲喋呤对粒单系白血病细胞既有明显的抑制作用(P<0.01),又有诱导分化作用。
In this study, bone marrow cells of patients with acute non-lymphocytic leukemia were cultured in diffusion chamber. The rats were divided into control group and medication group, the treatment group and control group were observed by light microscopy, counting colonies. The control group and the drug group containing the colony of the agar taken out by electron microscopy. The results showed that CFU-GM colonies from normal bone marrow were composed of mature cells: L-CFU colonies of leukemia bone marrow had distinct leukemic properties; methotrexate had both significant inhibitory effects on myelomonocytic leukemia cells (P <0.01), but also induced differentiation.
