目的研究腺苷A2B受体激动剂Bay60-6583对脓毒症模型小鼠的治疗作用。方法采用腹腔注射活金葡菌(108 CFU)制备小鼠脓毒症模型,观察Bay60-6583(2 mg/kg)、头孢曲松钠(25 mg/kg)单独或Bay60-6583与头孢曲松钠(2 mg/kg+25 mg/kg)联合给药对脓毒症模型小鼠的治疗效果,设正常对照组和感染组。采用ELISA试剂盒检测给药后小鼠血清中炎性细胞因子TNF-α、IL-1β和IL-6的含量,并使用血球计数板计数小鼠腹腔灌洗液中白细胞数。结果 Bay60-6583和头孢曲松钠单独给药均降低脓毒症模型小鼠死亡率,但联合给药组几乎完全消除小鼠死亡,与感染组或单独给药组相比较,差异有统计学意义(P<0.05);Bay60-6583单独或联合给药均明显降低小鼠血清中炎性细胞因子含量(P<0.05),但不改变小鼠腹腔灌洗液中白细胞数(P>0.05),而头孢曲松钠单独用药则略微降低血清中TNF-α、IL-1β和IL-6的含量(P>0.05)。结论 Bay60-6583通过抑制脓毒症小鼠炎性细胞因子表达发挥其治疗作用,与头孢曲松钠联合用药是合理的脓毒症治疗策略。
Objective To study the therapeutic effect of adenosine A2B receptor agonist Bay60-6583 on sepsis model mice. Methods The model of sepsis was induced by intraperitoneal injection of Staphylococcus aureus (108 CFU). The effects of Bay60-6583 (2 mg / kg), ceftriaxone sodium (25 mg / kg) alone or Bay60-6583 with ceftriaxone Sodium (2 mg / kg +25 mg / kg) combined treatment of septic mice model, the normal control group and infected group. The levels of inflammatory cytokines TNF-α, IL-1β and IL-6 in the serum of the mice after administration were detected by ELISA kit, and the number of white blood cells in the peritoneal lavage fluid of the mice was counted by using a hemocytometer. Results Both Bay60-6583 and ceftriaxone sodium alone decreased the mortality in septic mice, but the combined treatment group almost completely abolished the death of mice, compared with the infected group or the group administered alone, the difference was statistically significant (P <0.05). Bay60-6583 alone or in combination significantly reduced the levels of inflammatory cytokines in mice serum (P <0.05), but did not change the number of white blood cells in peritoneal lavage fluid of mice (P> 0.05) While ceftriaxone sodium alone decreased the levels of TNF-α, IL-1β and IL-6 in serum (P> 0.05). Conclusion Bay60-6583 exerts its therapeutic effect by inhibiting the expression of inflammatory cytokines in septic mice. Combined with ceftriaxone sodium is a reasonable treatment strategy for sepsis.
