目的探讨转化生长因子β1(TGF-β1)和NADPH氧化酶在糖尿病大鼠早期肾损伤中的作用,以及普罗布考对肾脏保护作用的机制。方法将30只大鼠随机分为正常对照组(NC)、糖尿病肾病(DN)组、普罗布考治疗组(DP),单侧肾切除后链脲佐菌素(STZ)35mg·kg-1腹腔注射诱导制作大鼠糖尿病肾病模型,用药8周后观察大鼠血糖(Glu)、血脂(TG及T-CH)、血肌酐(Scr)、尿白蛋白排泄率(UAER)、肾质量、肾肥大指数,以及肾病理改变并检测TGF-β1和p47phox的表达。结果 DM组和DP组的肾质量、肾肥大指数、T-CH都升高,DM组的UAER明显高于DP组;TGF-β1和p47phox的表达,NC组明显低于DN组和DP组,而DN组的表达明显高于DP组。结论普罗布考对糖尿病早期肾损伤有一定的保护作用,可能是通过改善机体氧化应激状态,减少细胞外基质,减少尿蛋白实现的。
Objective To investigate the role of transforming growth factor-β1 (TGF-β1) and NADPH oxidase in the early stage of renal injury in diabetic rats and the protective effect of probucol on kidneys. Methods Thirty rats were randomly divided into normal control group (NC), diabetic nephropathy (DN) group, probucol treatment group (DP), STZ 35mg · kg-1 The model of diabetic nephropathy was induced by intraperitoneal injection. After 8 weeks of treatment, the levels of Glu, TG and T-CH, Scr, UAER, kidney mass, kidney Hypertrophy index, and pathological changes of nephridial and to detect the expression of TGF-β1 and p47phox. Results The renal mass, renal hypertrophy index and T-CH in DM group and DP group were all increased. The UAER in DM group was significantly higher than that in DP group. The expression of TGF-β1 and p47phox in NC group was significantly lower than that in DN group and DP group. The DN group was significantly higher than the DP group. Conclusions Probucol may have a protective effect on early diabetic kidney injury, possibly through improving oxidative stress, decreasing extracellular matrix and reducing urine protein.
