目的建立RP-HPLC同时测定舒胸方中盐酸川芎嗪、阿魏酸、羟基红花黄色素A、人参皂苷Rg_1、人参皂苷Rb_1、人参皂苷Re、三七皂苷R_1的方法。方法以舒胸方为研究对象,采用RP-HPLC法,以Inertsil ODS-SP(250 mm×4.6 mm,5μm)为分析柱,乙腈-0.05%磷酸水溶液为流动相,梯度洗脱(0~45 min,15%乙腈;45~60 min,15%~20%乙腈;60~80 min,20%~38%乙腈;80~90 min,15%乙腈),检测波长为203 nm,柱温30℃,体积流量1.0 mL/min,进样量为20μL。结果 7种成分盐酸川芎嗪、阿魏酸、羟基红花黄色素A、人参皂苷Rg_1、人参皂苷Rb_1、人参皂苷Re、三七皂苷R1线性范围分别为0.101 0~1.364、0.0640~0.867 0、1.020~13.77、1.699~22.95、1.869~25.23、0.171 4~2.314、0.516 0~6.966μg/mL,线性关系良好,r值均在0.999 3~0.999 9,平均加样回收率在97.25%~103.52%,RSD均<3%。结论该方法简便、准确可靠,重复性好,分离度好,适用于同时测定舒胸方中主要7种有效成分。
Objective To establish a method for the simultaneous determination of ligustrazine hydrochloride, ferulic acid, hydroxysafflor yellow A, ginsenoside Rg_1, ginsenoside Rb_1, ginsenoside Re and notoginsenoside R_1 in Shu Cheng Fang by RP-HPLC. Methods Shu-cheng-fang was used as the research object. The mobile phase consisted of acetonitrile-0.05% phosphoric acid solution with gradient elution (0-45) using RP-HPLC method with Inertsil ODS-SP min, 15% acetonitrile, 45-60 min, 15% -20% acetonitrile, 60-80 min, 20% -38% acetonitrile, 80-90 min, 15% acetonitrile) at a detection wavelength of 203 nm with a column temperature of 30 ° C. , Volume flow 1.0 mL / min, injection volume 20 μL. Results The linear range of the seven components of ligustrazine hydrochloride, ferulic acid, hydroxysafflor yellow A, ginsenoside Rg_1, ginsenoside Rb_1, ginsenoside Re and notoginsenoside R1 were 0.101 0 ~ 1.364,0.0640 ~ 0.867 0,1.020 ~ 13.77,1.699 ~ 22.95,1.869 ~ 25.23,0.171 4 ~ 2.314,0.516 0 ~ 6.966μg / mL, the linear relationship was good, r values ?? were 0.999 3 ~ 0.999 9, the average recovery was in the range of 97.25% to 103.52% RSD <3%. Conclusion The method is simple, accurate and reliable, good repeatability, good resolution, suitable for the simultaneous determination of the main seven kinds of effective components Shu Shu.
