目的观察单独和联合使用全反式维甲酸(ATRA)和1,25-二羟维生素D3对大肠癌细胞株LoVo碱性磷酸酶(ALP)活性的变化的影响。方法用不同剂量的ATRA和1,25-二羟维生素D3单独或联合在体外作用于LoVo细胞2、4和6天,用脱氧胆酸钠溶解细胞,动力学方法测定ALP活性,紫外吸收法测定蛋白质含量。结果单独或联合使用ATAR和1,25-二羟维生素D3均可使LoVo细胞的肠型ALP活性逐渐升高,而以联合用药效果更好(P<0.05)。结论联合使用A-TRA和1,25-二羟维生素D3诱导LoVo细胞比单独用药效果更好,对于大肠癌的诊疗具要的指导意义
Objective To observe the effects of all-trans retinoic acid (ATRA) and 1,25-dihydroxyvitamin D3 on the changes of alkaline phosphatase (ALP) activity in colorectal cancer cell line LoVo. METHODS: Different doses of ATRA and 1,25-dihydroxyvitamin D3 were used alone or in combination to treat LoVo cells in vitro for 2, 4, and 6 days. Cells were lysed with sodium deoxycholate. ALP activity was determined kinetically and determined by ultraviolet absorption. Protein content. Results The use of ATAR and 1,25-dihydroxyvitamin D3 alone or in combination increased the intestinal ALP activity of LoVo cells, and the effect of combination therapy was better (P<0.05). Conclusion The combination of A-TRA and 1,25-dihydroxyvitamin D3 induces LoVo cells to be more effective than medication alone. It has guiding significance for the diagnosis and treatment of colorectal cancer.
