目的以羟丙基-β-环糊精(HP-β-CD)为手性选择剂,利用毛细管电泳法分离6种药物对映体。方法采用未涂层熔融石英毛细管柱,磷酸二氢钠缓冲溶液,分离电压20 kV,考察了背景电解质pH值、环糊精浓度、缓冲盐浓度对对映体分离的影响。结果在最优的电泳条件下,托吡卡胺、氨氯地平、尼卡地平、特布他林、盐酸曲马多和盐酸酚苄明的分离度分别为2.6、3.7、1.3、4.1、2.5和1.8。结论所建立的方法适于托吡卡胺、氨氯地平、特布他林、盐酸曲马多和盐酸酚苄明的对映体分离。
OBJECTIVE To separate the six enantiomers by capillary electrophoresis using hydroxypropyl-β-cyclodextrin (HP-β-CD) as a chiral selector. Methods Uncoated fused silica capillary column and sodium dihydrogen phosphate buffer solution were used to separate the membrane from each other at a voltage of 20 kV. The effects of background electrolyte pH, cyclodextrin concentration and buffer salt concentration on enantiomeric separation were investigated. Results The resolution of tropicamide, amlodipine, nicardipine, terbutaline, tramadol hydrochloride and phenoxybenzamine hydrochloride were 2.6, 3.7, 1.3, 4.1 and 2.5 respectively under the optimal electrophoresis conditions And 1.8. Conclusion The established method is suitable for the enantiomeric separation of tropicamide, amlodipine, terbutaline, tramadol hydrochloride and phenoxybenzamine hydrochloride.
