目的 荆介、紫苏叶为常用解表药 ,前期的实验研究已证实二者的挥发油部位具有良好的抗炎作用。为了进一步揭示它们的抗炎作用机理 ,研究了荆芥及紫苏叶挥发油对TNFα诱导的内皮细胞ICAM - 1表达的影响。方法 大鼠灌胃荆芥、紫苏叶挥发油后 ,分离含药血清 ,用细胞ELISA法观察TNFα激活 12、2 4h后内皮细胞表面ICAM - 1的表达。实验结果与空白血清组比较 ,同时以美洛昔康血清作为阳性对照。结果 TNFα(10 0 0kU/L)激活 12、2 4h ,ICAM - 1的表达极显著地增加 ;荆芥挥发油血清 (灌胃量为 0 2ml/kg、0 1ml/kg)能极显著抑制TNFα激活 12h后ICAM - 1的表达 ;紫苏叶挥发油血清 (灌胃量为 5 μl/kg、2 μl/kg)能显著抑制TNFα激活 12、2 4h后ICAM - 1的表达。结论 荆芥、紫苏叶挥发油对TNFα诱导的内皮细胞ICAM 1的表达有明显抑制作用 ,可能正是通过这种作用阻止了血管内皮细胞与白细胞的粘附 ,抑制白细胞向血管外移行 ,从而发挥抗炎作用
OBJECTIVE: Jing Jie and Perilla leaves are commonly used antiepidermal drugs. Previous experimental studies have confirmed that both volatile oils have good anti-inflammatory effects. In order to further reveal their anti-inflammatory mechanism, the effects of Nepeta and Perilla volatile oil on the expression of ICAM-1 in endothelial cells induced by TNFα were studied. Methods Rats were intragastrically perfused with essential oils from Nepeta and Perilla leaves, and serum containing drug was separated. The expression of ICAM-1 on the surface of endothelial cells after 12 and 24 hours of TNFα activation was observed by ELISA. The experimental results were compared with the blank sera group and meloxicam serum was used as a positive control. Results TNFα (100 kU/L) was activated for 12 and 24 h, and the expression of ICAM-1 was increased significantly; Schizonepeta volatile oil serum (0.2 mg/kg, 0.1 ml/kg) could significantly inhibit the activation of TNFα. The expression of ICAM-1 after 12h and the volatile oil serum of basil leaves (intragastric volume 5 μl/kg, 2 μl/kg) significantly inhibited the expression of ICAM-1 after 12 and 24 hours of TNFα activation. Conclusion The volatile oils from Nepeta and Perilla leaves inhibit the expression of ICAM 1 in endothelial cells induced by TNFα. This may prevent the adhesion of vascular endothelial cells to leukocytes and inhibit the leukocyte migration to blood vessels. Anti-inflammatory effect
