高温联合紫外线2~4个月连续暴露对大鼠血清白介素-21水平的影响

[目的]探讨高温联合紫外线2~4个月连续暴露对大鼠血清白介素-21(IL-21)水平的影响。[方法]选取体重为(200±10)g的健康成年SD大鼠112只,雌雄各半。随机分为对照组、高温组、紫外线组以及联合组(高温+紫外线),每组雌雄各14只,分笼饲养。在紫外灯(310 nm,72μW/cm2)和(38±1)℃高温环境中每天暴露2 h,连续暴露2个月和4个月后分别处死,采用酶联免疫吸附试验对大鼠血清IL-21进行检测。[结果]高温联合紫外线分别暴露2个月及4个月后,联合组雄性大鼠最终体重[(428.57±12.94)、(476.86±13.14)g],体重增长量[(183.29±11.74)、(306.86±13.81)g]明显低于对照组[最终体重:(493.57±17.12)、(587.00±11.53)g;体重增长量:(249.00±17.99)、(414.43±12.41)g](均P<0.05)。各组雌性大鼠体重差异均无统计学意义(P>0.05)。暴露2个月后,各暴露组无论雄性、雌性大鼠的脾脏系数和胸腺系数均无差异(P>0.05)。暴露4个月后,各暴露组雄性大鼠脾脏系数[高温组:(1.311±0.037)‰;紫外线组:(1.270±0.058)‰;联合组:(1.197±0.024)‰]均小于对照组[(1.410±0.026)‰](P<0.05);高温组雄性大鼠胸腺系数[(0.576±0.041)‰]小于对照组[(0.996±0.067)‰](P<0.05);而雌性大鼠的脏器系数均无差异(P>0.05)。相同暴露时间和暴露因素,雌、雄大鼠间血清IL-21水平差异均无统计学意义(P>0.05),故将雌雄大鼠合并分析。除2个月紫外线组[(487.69±15.21)ng/L]外,其他各暴露组[2个月:高温组(365.05±18.17)ng/L、联合组(278.66±11.70)ng/L;4个月:高温组(343.73±22.97)ng/L、紫外线组(264.45±23.96)ng/L、联合组(150.90±23.06)ng/L]大鼠血清IL-21水平均低于对照组[2个月(518.11±18.68)ng/L、4个月(582.18±28.90)ng/L](P<0.05);紫外线组和联合组大鼠暴露4个月时血清IL-21水平均低于暴露2个月时(P<0.01),无论暴露2个月还是4个月,联合组大鼠血清IL-21水平均明显低于高温组和紫外线组(P<0.05)。[结论]高温联合紫外线暴露对大鼠体重增长量、免疫器官重量以及血清IL-21水平均有一定的影响,且随着暴露时间的延长,血清IL-21含量呈降低趋势。 [Objective] To investigate the effect of continuous exposure of high temperature combined with UV light for 2 ~ 4 months on the level of serum interleukin-21 (IL-21) in rats. [Method] A total of 112 healthy adult SD rats weighing 200 ± 10 g were selected and divided into two groups, male and female. Randomly divided into control group, high temperature group, UV group and combined group (high temperature + UV), each group of 14 male and female, cage feeding. The rats were exposed to UV light (310 nm, 72 μW / cm2) and high temperature (38 ± 1) ℃ for 2 h every day, and were sacrificed 2 and 4 months after the continuous exposure. Serum IL -21 for testing. [Results] The final weight of male rats in the combined group [(428.57 ± 12.94), (476.86 ± 13.14) g], weight gain [(183.29 ± 11.74), ( 306.86 ± 13.81) g was significantly lower than the control group [final body weight: (493.57 ± 17.12), (587.00 ± 11.53) g; weight gain: (249.00 ± 17.99), (414.43 ± 12.41) g] ). There was no significant difference in weight between female rats in each group (P> 0.05). After exposure for 2 months, there was no difference in spleen coefficient and thymus coefficient between male and female rats in each exposure group (P> 0.05). After exposure for 4 months, the spleen coefficient of male rats in each exposed group [(high temperature group: (1.311 ± 0.037) ‰; ultraviolet group: (1.270 ± 0.058) ‰; combination group: (1.197 ± 0.024) ‰] (1.410 ± 0.026) ‰] (P <0.05). The thymus coefficient of male rats in high temperature group [(0.576 ± 0.041) ‰] was lower than that in control group [(0.996 ± 0.067) There was no difference in organ coefficient (P> 0.05). The same exposure time and exposure factors, male and female rats serum IL-21 levels were no significant difference (P> 0.05), so the male and female rats were combined analysis. Except for the 2-month UV group [(487.69 ± 15.21) ng / L], the other exposure groups [2 months: high temperature group (365.05 ± 18.17) ng / L and combination group (278.66 ± 11.70) ng / L; Month: Compared with control group, serum IL-21 levels in high temperature group (343.73 ± 22.97) ng / L, UV group (264.45 ± 23.96) ng / L and combined group (150.90 ± 23.06) ng / L] (518.11 ± 18.68) ng / L for 4 months (582.18 ± 28.90 ng / L) for 4 months (P <0.05). The levels of IL-21 in serum were lower than those in exposed group At 2 months (P <0.01), serum IL-21 levels in the combined group were significantly lower than those in the high-temperature group and the UV-light group at 2 and 4 months of exposure (P <0.05). [Conclusion] The combination of hyperthermia and UV exposure had certain effects on the weight gain, the weight of immune organs and the level of serum IL-21. With the prolongation of exposure time, the content of IL-21 in serum decreased.
