目的评价巢式PCR和荧光定量PCR方法在嗜肺军团菌气溶胶检测中的应用。方法于2012年8—10月在北京、南京、苏州和深圳四城市选取38家公共场所,使用带有虚拟浓缩器的Bio-sampler生物冲击式采样器采集气溶胶样品,分别应用巢式PCR和荧光定量PCR法检测样品中的嗜肺军团菌。结果共采集气溶胶样品165份。巢式PCR法测定嗜肺军团菌的阳性率为8.5%(14/165),荧光定量PCR法测定嗜肺军团菌的阳性率为26.1%(43/165),后者高于前者,差异有统计学意义(P<0.05)。结论采用荧光定量PCR法测定空气中嗜肺军团菌的灵敏度优于巢式PCR法。
Objective To evaluate the application of nested PCR and real-time quantitative PCR in the detection of Legionella pneumophila aerosol. Methods 38 public places were selected in Beijing, Nanjing, Suzhou and Shenzhen from August to October in 2012. Aerosol samples were collected using Bio-sampler biological impact sampler with virtual concentrator. Nested PCR and Detection of Legionella pneumophila in Fluorescent Quantitative PCR. Results A total of 165 aerosol samples were collected. The positive rate of Legionella pneumophila by nested PCR was 8.5% (14/165). The positive rate of Legionella pneumophila was 26.1% (43/165) by fluorescence quantitative PCR, which was higher than the former Statistical significance (P <0.05). Conclusion The detection of Legionella pneumophila in the air by fluorescence quantitative PCR is superior to nested PCR in sensitivity.
