以根癌农杆菌GV3101为介导,用茄红素-β-环化酶(lycopene bate cyclase-LYC-B)5′端反义基因对胡萝卜(Daucus carotaL.var.sativa)进行了遗传转化,经胚发生途径得到抗性植株。GUS检测和Southern杂交表明目的基因已整合到胡萝卜基因组中,Dot Blot显示目的基因在受体植株中已表达,茄红素和胡萝卜素检测结果表明,转基因愈伤组织茄红素含量高于胡萝卜素含量。
The Agrobacterium tumefaciens strain GV3101 was used to transform carrot (Daucus carota L.var.sativa) with the 5’-end antisense gene of lycopene b-cyclase-LYC-B, Resistant plants were obtained by means of embryogenesis. GUS detection and Southern hybridization showed that the target gene has been integrated into the carrot genome, Dot Blot showed that the target gene has been expressed in the recipient plant, lycopene and carotene test results show that the transgenic callus lycopene content higher than the carotene content.
