目的:比较研究8-异戊烯基柑橘素和柑橘素对体外培养大鼠股骨组织代谢活性的影响。方法:体外分离培养大鼠股骨组织,然后随机分为4组,其中3组分别给予雌二醇(终浓度为1×10-8mol·L-1)、8-异戊烯基柑橘素(终浓度为1×10-5mol·L-1)和柑橘素(终浓度为1×10-5mol·L-1),另外1组加生理盐水作为对照。分别在培养的第3、6、9、12、15和18 d测定大鼠股骨组织中碱性磷酸酶活性和钙含量,并采用RT-PCR检测骨保护素、Ⅰ-型胶原、Runx-2和骨形态发生蛋白基因的mRNA表达水平。结果:8-异戊烯基柑橘素、柑橘素和雌二醇均可提高股骨组织中碱性磷酸酶的活性,增加钙含量,并上调opg、bmp-2、collagen-I和runx-2 mRNA的表达水平;8-异戊烯基柑橘素活性强于柑橘素。结论:8-异戊烯基柑橘素和柑橘素均能改善体外培养大鼠股骨组织的代谢活性。
Objective: To compare the effects of 8-isopentenyl citrus and citrus on metabolic activity of rat femur in vitro. Methods: Femur tissue was isolated and cultured in vitro and then divided into 4 groups randomly. The 3 groups were given estradiol (final concentration of 1 × 10-8 mol·L-1), 8-isopentenyl citrus Concentration of 1 × 10-5mol·L-1) and citrus (final concentration of 1 × 10-5mol·L-1), the other group with saline as a control. The activities of alkaline phosphatase and calcium in rat femur tissue were measured on the 3rd, 6th, 9th, 12th, 15th, 15th and 18th day respectively. The expressions of osteoprotegerin, type Ⅰ collagen, Runx-2 And bone morphogenetic protein gene mRNA expression levels. Results: 8-isopentenyl citrus, citrus and estradiol all increased alkaline phosphatase activity, increased calcium content and up-regulated opg, bmp-2, collagen-I and runx-2 mRNA The expression level of 8-isopentenyl citrus was stronger than that of citrus. Conclusion: 8-isopentenyl citrus and citrus can improve the metabolic activity of rat femur tissue in vitro.
