目的 研究不同碘水平饮食对实验性自身免疫性甲状腺炎 (EAT)大鼠甲状腺的Th1型细胞因子IFN -γ和Th2型细胞因子IL - 10表达的影响 ,进一步探讨碘在自身免疫性甲状腺疾病 (AITD)发病中的作用机理。方法 雌性Lewis大鼠用低碘饲料和含不同浓度的KI水喂养 ,即分为低碘 (LI)、适碘 (NI)和高碘 (HI)组。EAT诱发组用Wis tar大鼠Tg免疫 ,诱发EAT的形成 ,观察大鼠甲状腺病理改变、血清中Tg自身抗体水平和甲状腺IFN -γ和IL -10mRNA的表达水平。结果 (1)EAT诱发的HI组大鼠甲状腺炎细胞浸润明显 ,接近重度炎症 ,NI组大鼠甲状腺为近炎症恢复期 ,而LI组呈轻度炎症。 (2 )EAT诱发组的HI、NI和LI大鼠血清中抗Tg自身抗体平均水平明显高于非EAT诱发对照组 ,P <0 0 1。 (3)EAT诱发组中的HI组大鼠甲状腺IFN -γ和IL - 10mRNA的表达 ,均明显高于非EAT诱发对照组 ,P <0 0 5。NI组大鼠甲状腺IL - 10mRNA和LI组甲状腺IFN -γmRNA的表达均高于非EAT诱发对照组 ,P <0 0 5。结论 EAT诱发组中的HI组大鼠甲状腺有较高水平的IFN -γ和IL - 10mRNA的表达 ,出现明显炎症和抗炎症反应 ;NI组大鼠仅Th2型细胞因子IL - 10较高水平表达 ,甲状腺炎症处于恢复期 ;LI组大鼠有较高水平IFN -γ的表达 ,甲状腺炎症反应较轻 ,其病程发展缓慢
Objective To investigate the effect of different dietary levels of iodine on the expression of Th1 cytokines IFN - γ and Th2 cytokines IL - 10 in thyroid of rats with experimental autoimmune thyroiditis (EAT), and to further explore the role of iodine in the pathogenesis of autoimmune thyroid diseases AITD) pathogenesis in the mechanism. Methods Female Lewis rats were fed with low iodine diet and different concentrations of KI water, which were divided into low iodine (LI), iodine (NI) and high iodine (HI) groups. The EAT-induced group was immunized with Wistar rat Tg to induce the formation of EAT. The pathological changes of thyroid, serum Tg autoantibodies and thyroid IFN-γ and IL-10 mRNA expression were observed. Results (1) Thyroiditis cells infiltrated by EAT in HI group were similar to those in severe inflammation. The thyroid in NI group was near inflamed and the mild in LI group. (2) The average level of anti-Tg autoantibodies in serum of HI, NI and LI rats in EAT-induced group was significantly higher than that in non-EAT-induced control group (P <0.01). (3) The expression of thyroid IFN - γand IL - 10mRNA in HI group in EAT induced group were significantly higher than that in non - EAT induced group (P <0.05). The thyroid IL - 10 mRNA in NI group and the thyroid IFN - γmRNA expression in LI group were higher than those in non - EAT group (P <0.05). Conclusion The expression of IFN - γ and IL - 10 mRNA in the thyroid of HI rats in EAT induced group shows obvious inflammation and anti - inflammatory reaction. Only the high expression of Th2 cytokine IL - 10 in NI rats , Thyroid inflammation in recovery phase; LI group rats have a higher level of IFN-γ expression, mild thyroid inflammation, the course of its slow development
