目的研究miR-152分子对JEG-3细胞中HLA-G表达水平的调控。方法在滋养细胞肿瘤细胞系JEG-3细胞中分别转染pre-miR-152(实验组),Cy3标记的pre-miRNA-control(阴性对照),空白对照(NC)为未转染的JEG-3细胞。转染48h、72h分别进行RT-PCR与Western blot实验检测细胞中miR-152、HLA-G mRNA与蛋白的表达水平。结果 RT-PCR结果显示在细胞中转染pre-miR-152分子后miR-152的表达显著增高(P<0.05);转染后HLA-G mRNA的表达水平在各组之间差异无统计学意义(P>0.05)。Western blot结果显示与对照组相比,实验组的HLA-G蛋白表达水平显著降低(P<0.05)。结论 miR-152可能在翻译水平抑制HLA-G的表达。
Objective To study the regulation of HLA-G expression in JEG-3 cells by miR-152. Methods The pre-miR-152 (experimental group), Cy3-labeled pre-miRNA-control (negative control) and blank control (NC) were transfected into the trophoblastic tumor cell line JEG- 3 cells. The expression of miR-152 and HLA-G mRNA and protein were detected by RT-PCR and Western blot respectively 48 h and 72 h after transfection. Results The results of RT-PCR showed that the expression of miR-152 was significantly increased in cells transfected with pre-miR-152 (P <0.05). There was no significant difference in the expression of HLA-G mRNA between the two groups Significance (P> 0.05). Western blot results showed that compared with the control group, the experimental group of HLA-G protein expression was significantly lower (P <0.05). Conclusion miR-152 may inhibit the expression of HLA-G at the translational level.
