目的进一步探讨叶酸和α-萘黄酮预防胎鼠腭裂发生的作用,并对比两者的效果。方法将孕鼠随机分为7组,每组8只,于受孕第10天(GD10)以四氯二苯二英(TCDD)28μg/kg灌胃;对照组以玉米油0.1 ml灌胃;叶酸组分别以叶酸15、10、5 mg/kg+TCDD 28μg/kg灌胃;α-萘黄酮分别以α-萘黄酮5 mg/kg、50μg/kg+TCDD 28μg/kg灌胃。受孕第17.5天(GD17.5)处死孕鼠,称量孕鼠及胚胎体重,记录活胎鼠数、腭裂数、吸收胎及死胎鼠数;并剪取胎鼠头部,解剖显微镜下观察。结果TCDD 28μg/kg所致胎鼠腭裂发生率为92.86%,对照组未见胎鼠腭裂形成。TCCD+叶酸15、10、5 mg/kg三组致胎鼠腭裂发生率分别为84.00%、73.08%、86.00%。α-萘黄酮5 mg/kg、50μg/kg+TCDD组胎鼠腭裂发生率分别为65.52%、74.00%。各实验组孕鼠体重增加量、活胎鼠体重及每窝平均胎数、每窝活胎数、死胎和吸收胎数与对照组及TCDD组相比,差异均无统计学意义。结论试验剂量的叶酸与α-萘黄酮均有一定的拮抗TCDD诱导胎鼠腭裂发生的作用,以叶酸10 mg/kg、α-萘黄酮5 mg/kg的剂量的拮抗作用较强;两者作用效果无明显差异,且对孕鼠及胎鼠的生长发育无明显影响。
Objective To further explore the role of folic acid and α-naphthalene flavone in preventing the development of fetal rat palate and to compare the effects of the two. Methods Pregnant mice were randomly divided into 7 groups (8 rats in each group). The pregnant rats were orally administered with TCDD 28 μg / kg on the tenth day of conception (GD10), while the control group was orally administered with 0.1 ml corn oil. Folic acid group were folic acid 15,10,5 mg / kg + TCDD 28μg / kg intragastric administration; α-naphthoflavone α-naphthoflavone 5 mg / kg, 50μg / kg + TCDD 28μg / kg gavage. Pregnant mice were sacrificed on day 17.5 (GD17.5). Body weights of pregnant mice and embryos were weighed. The number of live fetus, cleft palate and aborted fetuses were measured. The heads of fetal mice were dissected and observed under a dissecting microscope. Results The incidence of cleft palate in fetal rat with TCDD 28 μg / kg was 92.86%. No cleft palate was found in the control group. TCCD + folic acid 15,10,5 mg / kg three groups of fetal cleft palate incidence rates were 84.00%, 73.08%, 86.00%. The incidences of cleft palate in the 5-mg / kg α-naphthoflavone and 50 μg / kg + TCDD groups were 65.52% and 74.00%, respectively. Compared with the control group and the TCDD group, there was no significant difference in body weight gain, live body weight and average litter size per litter, live litter size per litter, stillbirth and aborted fetuses in each experimental group. Conclusions Both folic acid and α-naphthoflavone at certain doses can antagonize TCDD-induced cleft palate. The antagonistic effects of folic acid 10 mg / kg and α-naphthalene flavone 5 mg / kg are strong. No significant difference in the effect, and the growth of pregnant rats and fetal rats had no significant effect.
