目的采用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF/MS)联用技术对核桃楸皮进行分析,确定其主要化学组分。方法采用Waters Acquity UPLC BEH C18色谱柱(100 mm×2.1 mm,1.7μm),以0.1%甲酸水(A)-0.1%甲酸乙腈(B)为流动相梯度洗脱;质谱采用电喷雾(ESI)离子源,在正负离子模式下采集数据,通过Peakview 2.0/masterview1.0及Markerview1.2.1软件进行目标及非目标筛查化合物,依据精确质量数和同位素峰度比确定化合物分子式,通过对照品及数据库的二级谱图比对、裂解规律分析,结合已有文献报道,确定化合物结构式。结果鉴定或推断了核桃楸皮中44种主要化学成分,其中包括13个萘醌类化合物,3个二芳基庚烷类化合物,15个黄酮类化合物及13个其他类化合物。结论该方法快速、准确,为核桃楸皮的化学成分鉴定提供一种新的策略,主要化学成分的确定为质量评价指标选择及药理活性研究奠定了基础。
Objective To determine the main chemical components of walnut by using UPLC-Q-TOF / MS combined with ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry. Methods Waters Acquity UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) was used to elute the mobile phases with 0.1% formic acid (A) and 0.1% formic acid (B) Ion source, collecting data in positive and negative ion mode, and using both Peakview 2.0 / masterview1.0 and Markerview1.2.1 software to perform target and non-target screening compounds. The molecular formula of the compound is determined according to the exact mass number and isotope kurtosis ratio, Of the secondary spectrum comparison, analysis of the cracking, combined with the literature has been reported to determine the structural formula of the compound. Results The main chemical constituents of Juglans mandshurica were identified or inferred, including 13 naphthoquinone compounds, 3 diarylheptane compounds, 15 flavonoid compounds and 13 other compounds. Conclusion The method is rapid and accurate, which provides a new strategy for the chemical constituents identification of Juglans mandshurica. The determination of the main chemical components lays the foundation for the selection of quality evaluation indexes and the study of pharmacological activities.
