目的:探讨原癌基因C-myc和细胞凋亡抑制基因bcl-2产物在睾丸肿瘤中表达的意义及相关关系,了解细胞增殖与凋亡的特点。方法:采用免疫组化SP法对38例睾丸肿瘤和10例正常睾丸组织中的C-myc和bcl-2蛋白进行定量检测。结果:C-myc蛋白在睾丸肿瘤中阳性表达例数31/38,阳性细胞百分率为24.99%,正常睾丸组织阳性例数0/10,差异有高度显著性(P<0.01),但其表达与睾丸肿瘤病理分级、临床分期无关(P>0.05)。bcl-2蛋白的表达与肿瘤分级、分期密切相关(P<0.05),随恶性程度增加,其表达强度降低。C-myc与bcl-2的表达无显著相关性(r=0.33,P>0.05)。结论:检测睾丸肿瘤组织中bcl-2蛋白的表达可以作为睾丸肿瘤恶性程度和预后的估价性指标;C-myc蛋白的异常表达可能在睾丸肿瘤的发生、发展过程中起着促进作用。
Objective: To investigate the significance of the expression of the oncogene C-myc and the apoptosis-suppressor gene bcl-2 in testicular tumors and to understand the characteristics of cell proliferation and apoptosis. Methods: C-myc and bcl-2 protein in 38 cases of testicular tumor and 10 cases of normal testis were detected by immunohistochemical SP method. Results: The positive expression rate of C-myc in testes was 31/38, the percentage of positive cells was 24.99%, the positive number of normal testes was 0/10, the difference was highly significant (P <0.01) Testicular tumor pathological grade, clinical stage (P> 0.05). The expression of bcl-2 protein was closely related to tumor grade and staging (P <0.05), and decreased with the increase of malignant degree. There was no significant correlation between the expression of C-myc and bcl-2 (r = 0.33, P> 0.05). Conclusion: Detecting the expression of bcl-2 protein in testicular tumor can be used as a marker of malignancy and prognosis of testicular tumor. Abnormal expression of C-myc protein may play an important role in the development of testicular tumor.
