目的分析测定芜菁多糖中单糖的组成。方法采用水提醇沉法提取芜菁多糖,用H2SO4将多糖水解成单糖,1-苯基-3-甲基-5-吡唑啉酮衍生化,RP-HPLC分离检测。结果测得芜菁多糖是由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖和阿拉伯糖等单糖组成。6种单糖在25min内可很好分离,并对各种单糖进行了方法学考察。结论本法测定多糖的单糖组分操作简便、灵敏度高、结果准确可靠,可用作芜菁多糖的质量控制。
Objective To analyze and determine the composition of monosaccharides in the polysaccharide of turnip. Methods Polysaccharides were extracted by aqueous extraction and alcohol precipitation, the polysaccharides were hydrolyzed into monosaccharides by H2SO4, derivatized with 1-phenyl-3-methyl-5-pyrazolone and detected by RP-HPLC. The results showed that the polysaccharide of turnip is composed of monosaccharides such as mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose. Six kinds of monosaccharides can be well separated in 25min, and a variety of monosaccharides methodological study. Conclusion The monosaccharide composition of polysaccharide determined by this method is simple, sensitive, accurate and reliable, and can be used as quality control of polysaccharides.
