目的:探讨大肠癌细胞CD44V6的表达与其转移潜能之间的关系。方法:应用原位分子杂交、免疫组化、免疫荧光定量分析技术,从蛋白水平到分子水平、从定性到定量,分析不同转移能力人大肠癌细胞CD44V6的表达。结果:人大肠癌HT29、LoVo细胞系均有CD44V6变异体mRNA和蛋白表达。在mRNA和蛋白水平,高转移能力的LoVo细胞表达强度均高于低转移能力的HT29细胞。实验还发现:LoVo细胞系内不同的细胞之间表达CD44V6的强度不一致,部分LoVo细胞表达CD44V6明显增强。结论:CD44V6的表达与大肠癌细胞的转移潜能有关
Objective: To investigate the relationship between the expression of CD44V6 and its metastatic potential in colorectal cancer cells. Methods: The expression of CD44V6 in colorectal cancer cells with different metastatic potential was analyzed by in situ molecular hybridization, immunohistochemistry, and immunofluorescence quantitative analysis techniques from the protein level to the molecular level, from qualitative to quantitative. RESULTS: Human colorectal cancer HT29 and LoVo cell lines all had expression of CD44V6 variant mRNA and protein. At the mRNA and protein levels, the high metastatic ability of LoVo cells was higher than that of low metastatic ability of HT29 cells. The experiment also found that the intensity of the expression of CD44V6 was different between different cells in the LoVo cell line, and part of LoVo cells expressed CD44V6 was significantly enhanced. Conclusion: The expression of CD44V6 is related to the metastatic potential of colorectal cancer cells
