目的:观察创面愈合后不同时间应用90锶-90钇(90Sr-90Y)照射对防治瘢痕的作用。方法:建立兔耳瘢痕模型,9只兔随机分成A、B和C3组,每组15个创面。A组在创面愈后后第3天开始90Sr-90Y治疗,总剂量30Gy,每次剂量从2.5Gy开始,并逐渐增加至5.5Gy,隔日照射1次,分8次完成。B组在创面上皮化后第14天瘢痕增生后开始照射,剂量和使用方法同A组。C组为对照组,不应用90Sr-90Y照射。3组创面在B组完成90Sr-90Y照射后1周同时取材,应用HE染色观察形态学改变,Mas-son染色观察胶原纤维的分布,电镜学检查观察瘢痕组织超微结构的变化。结果:A组胶原纤维分布面积大幅低于B组和C组(P<0.05);A组电镜下成纤维细胞数量明显减少,体积相对较小,RER(粗面内质网)不发达,少许扩张成囊状,部分线粒体肿胀空泡化,核异染色质丰富,比例增加。结论:早期应用90Sr-90Y照射兔耳愈合创面有防治瘢痕增生的作用。
Objective: To observe the effect of 90Sr-90Y (90Sr-90Y) irradiation on the prevention and treatment of scars at different time after wound healing. Methods: Rabbit ear scars model was established. Nine rabbits were randomly divided into A, B and C3 groups, each with 15 wounds. Group A started treatment with 90Sr-90Y on the third day after wound healing, with a total dose of 30Gy. Each dose started from 2.5Gy and gradually increased to 5.5Gy. The irradiation was repeated every other day for 8 times. In group B, scars proliferated on the 14th day after wound epithelization, and the dose and usage were the same as those in group A. Group C as a control group, not 90Sr-90Y irradiation. The wounds of three groups were taken at the same time one week after 90Sr-90Y irradiation. The morphological changes were observed by HE staining. The distribution of collagen fibers was observed by Mas-son staining. The ultrastructure of scar tissue was observed by electron microscopy. Results: The distribution area of collagen fibers in group A was significantly lower than that in group B and group C (P <0.05). The number of fibroblasts in group A was significantly reduced, the volume was relatively small, and the RER (rough endoplasmic reticulum) Dilated into sacs, some mitochondria swelling vacuolar, heterochromatin abundance, the proportion increased. Conclusion: Early application of 90Sr-90Y irradiated rabbit ear healing wounds have the effect of preventing and treating scar hypertrophy.
