目的 :探讨热休克反应对卵巢癌细胞A2 780凋亡的影响。方法 :将体外培养的卵巢癌细胞A2 780分为非热休克组 (NHS)和热休克组 (HS) ,两组分别由三组浓度放线菌素D(Act-D)诱导。采用脱氧核苷酸末端转移酶介导的dUTP缺口末端标记法 (TUNEL)和3H -TdR掺入试验检测肿瘤细胞凋亡指数 (AI)及DNA合成情况。结果 :HS组比NHS组凋亡指数明显下降 (P <0 .0 1 ) ;cpm值明显升高 (P <0 .0 1 ) ,呈现剂量依赖效应。结论 :热休克反应对卵巢癌细胞A2 780经Act -D诱导凋亡具有抑制作用 ,此作用与Act-D呈剂量依赖效应。
Objective: To investigate the effect of heat shock reaction on the apoptosis of ovarian cancer cell line A2 780. Methods: The A2 780 ovarian cancer cell line was divided into non-heat shock group (NHS) and heat shock group (HS). The two groups were induced by three concentrations Actinomycin D (Act-D). Tumor cell apoptosis index (AI) and DNA synthesis were detected by deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) and 3H-TdR incorporation assay. Results: Apoptosis index of HS group was significantly lower than that of NHS group (P <0.01); cpm value was significantly increased (P <0.01), showing a dose-dependent effect. Conclusion: Heat shock response inhibits the apoptosis of ovarian cancer cell line A2 780 induced by Act-D, which is in a dose-dependent manner with Act-D.
