目的:研究苦远志中糖酯类化学成分及其抗氧化及抗黄嘌呤氧化酶活性。方法:采用D101大孔树脂、硅胶柱层析、Sephaclex LH-20、半制备高效液相色谱等多种方法分离纯化,并结合MS、NMR等波谱分析方法鉴定化合物的结构。利用ABTS方法和尿酸生成法分别测定其抗氧化和抗黄嘌呤氧化酶的活性。结果:从苦远志乙醇提取物中分离得到8个糖脂类化合物,分别鉴定为:蔗糖-6-苯甲酸酯(1)、sibiricose A6(2)、sibiricose A1(3)、glomeratose B(4)、sibiricose A3(5)、glomeratose A(6)、glomeratose D(7)、tenuifoliside C(8)。化合物4、7和8表现出较明显的抗氧化活性和抗黄嘌呤氧化酶抑制活性。结论:所有化学成分均为首次从该植物中分离得到,糖酯类成分在抗氧化与抗黄嘌呤氧化酶趋势上基本一致。
OBJECTIVE: To study the chemical constituents of sugar esters and their anti-oxidation and anti-xanthine oxidase activities in Bitterness Zhiyuan. Methods: The compounds were isolated and purified by D101 macroporous resin, silica gel column chromatography, Sephadex LH-20 and semi-preparative high performance liquid chromatography. The structures of the compounds were identified by MS, NMR and other spectroscopic methods. Anti-oxidative and anti-xanthine oxidase activities were determined by ABTS and uric acid generation, respectively. Results: Eight compounds were isolated from the ethanol extract of Bitterness Polygalaceae and identified as sucrose-6-benzoate (1), sibiricose A6 (2), sibiricose A1 (3), glomeratose B ), Sibiricose A3 (5), glomeratose A (6), glomeratose D (7), tenuifoliside C (8). Compounds 4, 7 and 8 showed more pronounced antioxidant activity and anti-xanthine oxidase inhibitory activity. Conclusion: All the chemical components were isolated from this plant for the first time. The components of sugar esters were almost the same in anti-oxidation and anti-xanthine oxidase.
