目的观察口服雷奈酸锶(strontium ranelate,SR)与胶原肽(collagen peptides,CP)对尾吊(tail supended,TS)大鼠骨丢失的抑制效应。方法雄性SD大鼠24只随机平均分为3组:对照组(CN),尾吊模拟微重力组(TS),模拟失重同时给予SR和CP各750 mg/kg灌胃组(SR&CP),各组实验期均为28 d。采用全自动生化分析仪检测碱性磷酸酶(alkaline phosphatase,ALP)含量,放射免疫法检测骨钙素(osteocalcin,OC)含量;Micro-CT分析骨微结构;Masson染色观察骨胶原纤维;双能X线(dual energy X-ray absorptiometry,DXA)检测股骨骨密度(bone mineral density,BMD)。结果 TS大鼠补充SR和CP显示骨保护作用:抑制TS诱导的血清ALP和OC含量的降低,改善骨微结构;显著抑制TS组大鼠股骨BMD的降低。结论口服SR和CP能明显抑制TS大鼠的骨丢失,从而为微重力环境下骨丢失的防治提供了实验依据。
Objective To observe the inhibitory effects of oral administration of strontium ranelate (SR) and collagen peptides (CP) on bone loss in tail supended rats. Methods Twenty-four male Sprague-Dawley rats were randomly divided into 3 groups: control group (CN), tail microtissue (TS) tail suspension, simulated weightlessness (SR) The experimental period was 28 days. The content of alkaline phosphatase (ALP) was detected by automatic biochemical analyzer. The content of osteocalcin (OC) was detected by radioimmunoassay. Micro-CT was analyzed by Micro-CT. Collagen fibers were observed by Masson staining. X-ray (dual energy X-ray absorptiometry, DXA) was used to detect the bone mineral density (BMD). Results TS and SR supplementation showed osteoprotection: the decrease of serum ALP and OC levels induced by TS and the improvement of bone microarchitecture were observed. The reduction of femoral BMD in TS rats was significantly inhibited. Conclusion Oral administration of SR and CP can significantly inhibit bone loss in TS rats, which provides an experimental basis for the prevention and treatment of bone loss under microgravity.
