目的 探讨肿瘤坏死因子α(TNFα)抗体在大鼠急性出血坏死性胰腺炎时对脑组织损害的保护作用,从而为胰性脑病的治疗提供实验依据。方法 将60只SD大鼠随机分为正常对照组,急性出血坏死性胰腺炎诱导组和TNFα抗体处理组,经胰胆管逆行注射5%牛磺胆酸建立大鼠ANP模型,测定血的TNFα水平,12h后处死动物,检查胰腺组织形态学变化,并检测脑组织含水量、脑微血管内白细胞聚集及附壁现象。结果 经TNFα抗体处理后,血清中TNFα水平、脑组织含水量、脑微血管内白细胞聚集及附壁现象显著降低(P<0.05),胰腺组织病理损害程度减轻。结论 TNFα抗体可以减轻急性出血坏死性胰腺炎时脑损害的发生和发展。
Objective To investigate the protective effect of tumor necrosis factor α (TNFα) antibody on brain tissue injury in rats with acute hemorrhagic necrotizing pancreatitis, and to provide experimental evidence for the treatment of pancreatic encephalopathy. Methods Sixty Sprague-Dawley rats were randomly divided into normal control group, acute hemorrhagic necrotizing pancreatitis induction group and TNFα antibody-treated group. Rat model of ANP was established by retrograde injection of 5% taurocholic acid into the pancreaticobiliary duct. The levels of TNFα After 12 hours, the animals were sacrificed and the morphological changes of the pancreas were examined. The water content of brain tissue, the aggregation of leucocytes in the capillaries and the phenomenon of attachment were observed. Results After treatment with TNFα antibody, the level of TNFα in serum, the content of water in brain tissue, the aggregation of leukocytes in cerebral microvessels and the phenomenon of adherent cells were significantly decreased (P <0.05), and the degree of pathological damage in pancreatic tissue was relieved. Conclusion TNFα antibody can reduce the occurrence and development of brain damage in acute hemorrhagic necrotizing pancreatitis.
