以限制性内切酶HpaⅡ/MspⅠ消化、Southernblot方法,分析22例进展期胃癌手术标本的癌灶、癌旁和外周正常组织细胞内c-myc、c-Ha-ras癌基因位点的DNA甲基化状态,并与其组织病理学变化相比较。结果发现1例正常组织、13例癌旁和10例癌灶细胞c-myc基因和自显影较清楚的10组标本中的4例癌旁、5例癌灶细胞内c-Ha-ras基因呈低甲基化状态。以上改变与肿瘤大体形态、组织类型、癌细胞分化程度、淋巴结转移及癌细胞浸润深度等病理学改变无明显相关性。
The DNA methylation of c-myc and c-Ha-ras oncogene sites in the tumor foci, paracancerous and peripheral normal tissues of 22 advanced gastric cancer surgical specimens were analyzed by restriction enzyme HpaII/MspI digestion and Southern blot analysis. The status is based on and compared with histopathological changes. The results showed that the c-Ha-ras gene was found in 4 cases of paracancerous and 5 cases of foci in 10 specimens of 1 normal tissue, 13 paracancerous and 10 foci with c-myc gene and autoradiography. Hypomethylation state. The above changes were not significantly related to pathological changes such as gross morphology, histological type, degree of cancer cell differentiation, lymph node metastasis, and depth of cancer infiltration.
