目的探索脂质体、CpG寡核苷酸(ODN)以及Hepa 1-6肝癌细胞裂解物联合应用治疗肝癌的免疫效果。方法建立C57BL/6小鼠肝癌模型,以PBS、ODN+Hep(肝癌细胞Hep)、Lip(Lipofectamine脂质体)+HDN+Hep荷瘤鼠背部皮下注射,各组小鼠肝脏HE染色,FCM检测小鼠脾脏CD3-FITC、CD4-FITC、CD8-FITC、NK1.1-FITC的组成,ELISA法测免疫小鼠血清中IFN-γ、IL-4水平。结果脂质体包裹CpG ODN+Hepa 1-6肝癌细胞裂解物中CD3-FITC、CD4-FITC、CD8-FITC、NK1.1-FITC的体外杀伤活性以及荷瘤小鼠血清中IFN-γ、IL-4水平均显著高于CpG ODN+Hep组以及PBS组。结论脂质体包裹CpG ODN+Hep能显著提高机体的免疫功能,尤其是特异性细胞免疫功能。
Objective To explore the immunological effects of liposomes, CpG oligonucleotides (ODNs) and Hepa 1-6 hepatoma cell lysate in the treatment of liver cancer. Methods C57BL / 6 mice model of hepatocellular carcinoma was established. The mice were injected subcutaneously with PBS, ODN + Hep (Hep), Lip (Lipofectamine liposome) + HDN + Hep mice. The spleen CD3-FITC, CD4-FITC, CD8-FITC and NK1.1-FITC were detected by ELISA. The levels of IFN-γ and IL-4 in serum were detected by ELISA. Results The in vitro cytotoxic activity of CD3-FITC, CD4-FITC, CD8-FITC and NK1.1-FITC in the liposome-encapsulated CpG ODN + Hepa 1-6 hepatoma cell lysate and the serum IFN- γ, IL -4 levels were significantly higher than CpG ODN + Hep group and PBS group. Conclusion The encapsulation of CpG ODN + Hep by liposomes can significantly improve the immune function, especially the specific cellular immune function.
