本文检测子宫内膜癌两种肿瘤抑制基因,发现p53和视网膜母细胞癌(Rb)基因有高频率变异并检测候补转移抑制基因nm23H_1的状况。作者在72例典型子宫内膜增殖症患者和内膜癌患者中,经子宫切除术立即取活检标本用福尔马林固定,石蜡包埋,免疫组化检测p53基因状况。取10例子宫内膜癌的冰冻组织标本提取DNA,用nm23-H_1,cDNA特异探针杂交法,检测nm23基因的杂合丢失(LOH)状况。取10例子宫内膜癌和2例良性子宫内膜癌患者冰冻标本,进行免疫反应,观察视网膜母细胞瘤蛋白的典型定量来评估Rb基因状况。
In this study, we detected two tumor suppressor genes of endometrial cancer and found that there is a high frequency mutation of p53 and retinoblastoma (Rb) genes and the detection of the candidate metastasis suppressor gene nm23H_1. In 72 cases of endometrial hyperplasia patients and endometrial cancer patients, biopsy specimens were taken immediately after hysterectomy using formalin fixed, paraffin-embedded, immunohistochemical detection of p53 gene status. DNA was extracted from 10 frozen tissue specimens of endometrial carcinoma, and the hybridization loss (LOH) of nm23 gene was detected by nm23-H_1 cDNA hybridization. Frozen specimens from 10 patients with endometrial carcinoma and 2 patients with benign endometrial carcinoma were immunized and observed for the typical quantification of retinoblastoma protein to assess the status of Rb gene.
